چکیده

Abstract Background Despite the increased morbidity of ulcerative colitis (UC) in developing countries, available treatments remain unsatisfactory. Therefore, it is urgent to discover more effective therapeutic strategies. Pulsatilla chinensis was widely used for treatment inflamed intestinal diseases including UC thousands years China. Anemoside B4, most abundant triterpenoid saponin isolated from P. , exerts anti-inflammatory and antioxidant effects may be active compounds, which responsible effects. However, mechanism how anemoside B4 executes its biological functions still elusive. Methods Here, we 2, 4, 6-trinitrobenzene sulfonic acid (TNBS)-induced rat model evaluate effect B4. Blood samples rats were collected hematology analysis. The inflammation-associated factors investigated by enzyme-linked immunosorbent assay (ELISA). Cell proliferation apoptosis determined with EdU cell TUNEL assay. proteins regulated identified label-free quantitative proteomics. significantly down-regulated verified Western blotting mRNA expression analyzed real-time RT-PCR. Results results showed that ameliorated TNBS-induced symptoms, tissue damage, inflammatory infiltration, pro-inflammatory cytokine production, slowed colon. Quantitative proteomic analyses discovered 56 altered rats. These mainly clustered tricarboxylic (TCA) cycle respiratory electron transport chain. Among proteins, S100A9 one associated NF-κB MAPK signaling pathways pathogenesis UC. Further experiments revealed suppressed downstream genes TLR4 In vitro, could inhibit pathway induced recombinant protein human epithelial Caco-2 cells. Moreover, inhibits neutrophils recruitment activation colon TNBS. Conclusions Our demonstrate prevents inhibiting through deactivating S100A9, suggesting a promising candidate colitis.

Imiquimod-induced CCR9 Ameliorates murine TNBS Colitis

AIMS To investigate whether Imiquimod (IMQ) as TLR7 ligand protects mice from colonic inflammation and the mechanisms underlying in such immunoregulatory conditions. METHODS: Murine colitis was induced to Balb/c mice by administration of trinitrobenzene sulfonic acid (TNBS) with or without daily intraperitoneal administration of IMQ. Colitis was evaluated by body weight decreases and by histolo...

Glutamine relieves oxidative stress through PI3K/Akt signaling pathway in DSS-induced ulcerative colitis mice

Objective(s): Ulcerative colitis (UC) is a kind of complex immune disease, and a major cause of destruction of intestinal barrier and oxidative stress in this field. In this paper, glutamine (Gln) was believed to offer protection against oxidative stress injury in colitis mice.Materials and Methods: Thirty mice were randomly assigned int...

Sijunzi Decoction attenuates 2, 4, 6-trinitrobenzene sulfonic acid (TNBS)-induced colitis in rats and ameliorates TNBS-induced claudin-2 damage via NF-κB pathway in Caco2 cells

BACKGROUND SijunziDecoction (SJZD) is a traditional Chinese medicine prescription used to treat the diseases of gastrointestinal tract since ancient times. The objective of this study was to investigate the protective effects of SJZD on TNBS-induced colitis in rats and TNBS-damaged Caco2 cells. METHODS The rat colitis model was induced by 2, 4, 6-trinitrobenzene sulfonic acid (TNBS). SJZD (2....

Chrysin ameliorates chemically induced colitis in the mouse through modulation of a PXR/NF-κB signaling pathway.

Targeted activation of pregnane X receptor (PXR) in recent years has become a therapeutic strategy for inflammatory bowel disease. Chrysin is a naturally occurring flavonoid with anti-inflammation activity. The current study investigated the role of chrysin as a putative mouse PXR agonist in preventing experimental colitis. Pre-administration of chrysin ameliorated inflammatory symptoms in mous...

Kalopanaxsaponin B Ameliorates TNBS-Induced Colitis in Mice

The stem-bark of Kalopanax pictus (KP, family Araliaceae), of which main constituent is kalopanaxsaponin B, has been used for asthma, rhinitis, and arthritis in Chinese traditional medicine. To clarify anticolitic effect of KP, we examined anti-inflammatory effect of KP extract and kalopanaxsaponin B in lipopolysaccharide (LPS)-stimulated peritoneal macrophage and 2,4,6-trinitrobenzene sulfonic...
